To the best of my knowledge, sequencing is the preferred methodology for detecting SNPs when facilities are available. RFLP is a tedious and labor-intensive method, and restriction digestion may not be perfect. What about HRM analysis? What is the opinion of experts in this field?
Yeah, several studies have already suggested HRM as an alternative tool to DNA sequencing showing that HRM is a convenient, rapid and cost-efficient 'screening test' to detect the presence/absence of SNPs/mutations in a chosen DNA sequence. Whenever SNPs presence is indicated, DNA sequencing might be performed as a 'confirmatory test', when necessary, to 'confirm' and 'identify' such known/novel sequence variations encountered.
Droplet digital PCR is here - ... it may be helpful as well... but I'm not sure how cost-effective it is at this point.
