Hello Every one,

I need your suggestions regarding DMSO use for in vitro culture media of mouse embryo (KSOM media). I have planned an experiment where I have to use an antioxidant compound in mouse embryo culture media (KSOM media). The compound is soluble in DMSO, but I felt that after dissolving compound in DMSO and using the same compound in KSOM media retarded embryo development even in a very small concentration.

Kindly let me know if any one have used DMSO for any other chemical in KSOM media,

and is DMSO is recommended for Mouse culture media? or can any one suggest a less harm organic solvent which i can used in KSOM media?

Secondly from last one month my in vitro mouse embryo development is very low, and I found that the day5 mouse blastocyst development rate is very low. To find the reason of low blastocyst development I did the following

1. Freshly prepared PMSG and HCG hormone injection to improve ovulation and fertilization

2. Doing PMSG and HCG myself to minimize the error (Ovulation and embryo retrieval is fine)

3. Freshly prepared the enzyme Hyaluronidase

4. Prepared fresh stock of KSOM media

I also found that the incubator has polluted and found some yellow spot of fungus and just try to clean with tissue (the exposed internal part but not properly cleaned through technician)

Kindly give me your suggestions and recommendations of DMSO in KSOM media

Secondly what else I can do to improve blastocyst development to the normal level?
