For a human biomontoring study, I am planning to collect blood samples in EDTA tubes, store at -20°c for 5 days, and then transport in ice box to the lab where the samples will be deeply frozen (-80 °c) until analysis time by comet assay. Considering this procedure being without using cryopreservative or prior isolation of PBMC, anyone has an idea about the possibility of success for this procedure?

Yes, it's possible. The only thing the quality of slides will be not so good as slides prepared from fresh blood.

Surely. it is possible. I am working on short term preservation of sturgeon fish in Iran.I encountered with sperm DNA damage in a time dependent manner from day 0 up to day 5.It is a part of my PhD study. In your research, I think the DNA damage will be presented so, but be sure that the results from day to day will be surprisingly different from the day 0 afterwards. But, don't mind, because you can compare all samples in one specific day, for example day 5, only.
Remember, comet assay is a very wide and common study for cell toxicology among variety of tissues.For reliable comparison, just the situations of treatments should be unified.
