I'm going to use the B16F10 as a EMT model for my drug's anti-tumor study. However, rare information has been found on Internet for the EMT induction of B16F10. I wonder what's the best way to induce the EMT of B16F10 and if it's any phenotype changes while the EMT is happening.

Yeah B16F10 is a good model as it replicates really fast. But for better study try using any aggressive human cancer cells such as MDAMB231 or MCF7 or any other because at the end of the day, we are developing ways to combat human cancer.

I'm sorry I have to ask because I probably might have misunderstood the question. You want to induce EMT? B16-F10 are not really epithelial. Especially the phenotype isn't really one of either, neither epithelial nor mesenchymal. Beside the argument that human cancer could be more interesting, there is another aspect that could be convincing to use another cell panel. MCF-10A cells are really epithelial, MDA-MB-436 cells are cancerous but low metastatic. MDA-MB-231 are high metastatic (mesenchymal phenotype) cell lines with a visible differences in phenotype. If your drugs are anti-tumor drugs, do you want to induce an MET?
