I am going to start NSC34 cell culture and I need to know what precaution should I take before start. I am planing to do some flow experiment and transfection. Please help me out.
Concerning the transfection part of your question, I would like to offer you the possibility to try our Magnetofection*** based reagent NeuroMag.
initially developped for primary neurons, NeuroMag was demonstrated by many to be highly efficient on any type of primary neurons (hippocampal, cortical, motor neurons, amygdal...) and also on many neural cells of whom NSC34.
you can refer to the following papers for more information:
Finelli MJ., Hum Mol Genet. 2015 Mar 19. pii: ddv104.
Siddharthan V. PLoS One. 2014 Sep 2;9(9):e106535.
I am pretty sure NeuroMag will be of high importance for your transfection experiment, would you like to try it, please do not hesitate to contact me via researchgate or directly at : [email protected].
good luck with your experiment,
Cedric
MAGNETOFECTION
*** The Magnetofection technology uses a magnetic field to attract and concentrate complexes of magnetic nanoparticles and nucleic acid onto the cell surface as demonstrated by Grześkowiak BF et al, Pharm Res. 2014 Jul 18. (http://www.ncbi.nlm.nih.gov/pubmed/25033763).
Has anybody here tried out culturing these cells in serums other than FBS? I used New born calf serum and I see growth rate is very low compared to N2a for instance.
