Actually, I m trying to prepare hydrogel using fish collagen, rat tail collagen, and bovine collagen to check the cell adhesion and viability. fish and rat tail collagen nicely formed as a hydrogel when I mix with NaOH. but bovine collagen forms clumps. I could not find the exact PH for this bovine collagen hydrogel formation!!!!!

The pKa of the carboxylic acid and amine groups of collagen being respectively 4.1 and 9.3, below a pH of 4, the protonation of the amide groups leads to the generation of repulsive electrostatic forces that ensure the solubilization of collagen. Bovine collagen can then form physical hydrogels because the assembly of collagen fibers is governed by electrostatic and hydrophobic interactions regulated by pH, temperature and ionic strength. In order to obtain hydrogels from a solution of bovine collagen dissolved in NaOH, the pH value of the solution must be around 10.2.

Dear Induja, is it important to use hydrogel based on bovine collagen for the cell adhesion and viability studies? We undertake R&D and manufacturing of cross-linked hydrogels for biomedical applications, such as advanced wound healing. We do not use any animal derived chemicals or intermediates for making the hydrogels.
