I wanted to isolate these microorganisms from a commercial product and I did it in 37 and 30 C anaerobically and aerobically. The experiment was a failure. I bought these microorganisms from DSMZ and I want to grow them in M17 Broth. Is this a good idea? I know that L. lactis is grown at 30o C and S. thermophilus at 37o C anaerobically. Am I right???
You're right. Both L. lactis and S. thermophilus grow in M17; lactis at 30°C with glucose 1%, and Sthermophilus at 37 or 42°C with lactose 1% (its preferential carbon source, see Ben Yahia et al, 2012, Beneficial Microbes), without stiring (S. thermophilus is more sensitive to O2 than lactis is; gently homogenize liquid cultures). For solid medium cultures, you can use "anaerobic" conditions (Anaerocult, Merck). It will work!
