I was wondering if anybody is aware of a method I could use in mice to blunt PKA and/or MAPK signaling in adipose tissue. I have tried i.p. administration of H-89, which did not have any effect (even though I am not sure about which would be an appropriate control to test whether the signaling is blunted).
I would be happy about any suggestion!

Hi Karin, i think in vivo models are extremely difficult to get around when you have to focus on one cell type. The adipose tissue is dispersed along the abdominal cavity and also have less blood supply as compared the other organs. Secondly the appropriate tests for the signaling cascade of the adipocute should be adiponectin.

Adiponectin? I am interested in Brown adipose tissue, which is activated by norepinephrine and the subsequent activation of the PKA pathway.
So far I could not inibit this activation at the stage of PKA in vivo.

uncoupling protein 1 (UCP1), a mitochondrial protein I think should be the appropriate control to see whether the PKA signaling has been compromised or not.

thanks, in fact in vitro we could see that PKA Inhibition blocked Ucp1 Expression, however we want to look upstream of Ucp1, we want to see if some genes are regulated under the control of the PKA pathway, so measuring Ucp1 will not give me any Information other than a control of any potential Inhibitor i would find and apply.
