I have 3 treatment groups plus a control set of samples that have been assessed for protein output comparison via 2D gel electrophoresis. I need to reliably identify which spots are significantly up/down regulated between the groups for downstream MS analysis. Ludesi Redfin Solo was used for image quantification of spot volumes with predicted ANOVA score between the 4 groups highlighting some significant, but is it then necessary to also compare all treated vs control as well as each group to control instead of all 4 groups together or is there a more appropriate post-hoc test for this?

I dont know what spot volumes is. But if it is a quantitative normal variable, then you can employ ANOVA. Please note that in ANOVA, the hemogeneity (equality) of variances in groups is necessary. For assessing this, you can check "hemogeneity of variances" in Options menue in SPSS. If data are nonnormal or have different variances in the groups, Kruskal-Wallis H tests is the best choice.
Ruth, ANOVA tests if a significant difference exists in at least one pair among at least 3 pairs. It does not test the significance in all pairs. If significance is detected, the researcher then proceeds to multiple comparison test (to test if there is significant difference in every pair).
