I have a big problem with INS-1E cell line. I thawed 2 stocks of INS-1E cells (frozen in April 2010 in N2) and they simply do not adhere to the flask surface. Does anyone work with this cell line? Any advice? Thank you very much :))
I thawed twice INS-1E cells and I can say that they attach&recover very slowly. They are quite fragile and sensitive to DMSO, I guess. The best thing is to put a lot of medium, leave them alone for two-three days and don't pass them until they reach reasonable confluence. You can change medium after they are attached but I prefer not wash with PBS in the first passages. Also try to use Falcon flasks, which seems better for them.
I'm thawing INS cells for the third time in these days also.. I thawed them on Tuesday and only now they are starting to attach.. if they don't attach in a week, simply thaw another stock :)
