Yes, you can. The extra wells can be removed, sealed and they go back into the freezer. But, the biggest challenge we had faced while doing so was with the reagent. Especially which you are using TMB substrate which is light sensitive and once opened is almost difficult to reuse as it would change colour and give inappropriate colour.
If your kit has strip wells, yes. If a solid plate, no. With a minimum of 16 wells for your standards, you are already @ 76 wells, so you'll have two strips left over. I used to wrap them back up with the dessicant and seal with tape. Be sure to use the leftover strips with the same lot. If you are mixing substrate you won't be able to reuse the mixed portion, but my experience with both the premixed and A + B solutions is that they are pretty stable if you treat them as if they were sterile and you were trying to keep them that way. Overkill maybe=) And protect them from light in the refrigerator. I ran insulin ELISAs this way for years, only throwing out strips/reagents when going to a new lot. Never had a problem.
