In our lab, to block internalization and endocytosis of EGFR, an inhibitor solution of 2 mM of sodium fluoride (NaF), 10 mM of sodium azide (NaN3) and 5 mM of 2-deoxy-D-glucose is used.
Someone started using it many years ago and everyone else just followed. Nobody seems to know why this particular composition is used. Why are these particular salts used and how do they actually work? Are there better alternatives?
