miRNAs are regulated by both transcriptional dependent and transcriptional independent ways . In transcriptional dependent pathways lot of transcriptional factors are known to be involved in transcribing miRNA genes ultimately leading to over expression of miRNAs. In transcriptional independent pathways the processing of primiRNA to premiRNA and premiRNA to matured miRNA are either enhanced or diminished depending on the signaling molecule/trancription factors/ receptors interaction with either drosha or dicer which are known to play key role in processing the miRNAs. During normal healthy conditions these pathways are well regulated but during unphysiological conditions like cancer/ metabolic syndrome etc.. where.the cell signalling is deregulated. so these highly expressed signaling molecules either by directly interacting with miRNA processing machinery(drosha/ dicer) or indirectly controlling the transcriptional machinery leads to up regulation of miRNAs. These upregulated miRNAs now targets all the important genes required for proper functioning of the physiological system either by translation repression or mRNA degradation there by leading to a diseased condition.
