I've been working on CdCl2 toxicity on PC12 cells for months but every time I get different results with the same IC50 in the same conditions as before: RPMI 1640 - 6000 cells/well - 100 microL/well
Does anybody know how can I get more repeatable and dependable outcomes?? Is there anything I should try or change??

Hi Mersedeh, I assume you're interested in the toxicity of Cd2+ and not necessarily of CdCl2, so perhaps you could switch to Cd(NO3)2 if that is more soluble. Also carefully check the pH (before and after the end of the test; use a micro pH meter for a measurement in the well or alternatively collect the medium from several replicate wells to achieve a sufficient volume to measure pH) and composition of your test medium and consider if there are constituents with which Cd could form a complex and be less bioavailable. Set the pH as low as possible to keep Cd in solution, but within the tolerated range for your cells so that they are not compromised in their performance. Issues with solubility and complexation are often the problem in working with metals. Hope this helps.
