In literature, why does the viral source for the viral infection of human cells always come from DENV-infected mosquito cells (e.g. C6/36 cells)?
Does that mean that mosquito factors can not be absent in DENV infection of human cells in vitro?

We know than in vivo it is necessary an enhancement of the DENV in the mosquito cells before it can infect vertebrate cells (the example is in human cells). In the mosquito there is a process called extrinsical incubation period, during this time there is replication of viral particles before they can go to the mosquito salivary glands to be released during the mosquito bite (during its blood feeding). However there are reports of transmission through blood products (transfusions) depending on the concentration of viral particles

This clinical experience leads to think that under special conditions the
virus can replicate in new cells without a mosquito mediating this transmission. It is worthy to explore this possibility since nothing is absolute in medicine.
Hi,
in an in vivo situation, dengue needs an intermediary vector because it cannot be transmitted directly from human to humans. In an in vitro situation, mosquito cells are used only to increase the titre of the replicating virus to be used for further experiments. Apart from that, a dengue culture supernatant infected from one type of mammalian cell can infect the other mammalian cell too if its titre is high or in an appropriate amount.
DENV can be used in human cells for in-vitro studies, however virus production is low and you will always need new viral stocks from C6/36 cells.
