I'm using a Nikon Eclipse microscope, and I wanted to make sure that images from different objectives (e.g., 10x, 40x) would be centered at the same stage position.  Strangely, they were centered in one direction (y), but not in the other (x).  The fact that x and y behave differently is very confusing.

I attached a graph to show what I mean.  I centered the scope on a particular cell at 10x magnification.  The image size was 2048x2048, so the center was at ~1024.  As I switched to 20x, 40x, and 60x magnification, the cell appeared more and more off-center, but only in one direction.

Does anyone know what might be happening here?  Maybe there is something misaligned which I need to fix?  Thanks!
