ONB is a very rich and complete medium, it contains yeast extract and peptione. Both contain histidine. No extra addition needed. Yet be sure to wash your cells for the next steps in the protocol because you will otherwise contaminate your test with histidine seemingly giving high revertant numbers in the controls.

ONB is an enriched media. It has beef extract and peptone which contains all the amino acid including histidine apart from other nutrients.
