Any one please help me to get a protocol for immunofluorescence staining of streptavidin coated particles bind to biotin fluorescent coated. I couldn't find an article related to that.
I normally add biotin from the same species as the primary antibody (concentration 1:500 for 1-2 hours Room Temp, then wash) and then add streptavidin conjugated to a fluorophore (concentration 1:250-1:500, 1-2 hours, wash). You may want to use an endogenous biotin blocking kit prior to staining. Also, a block/permeablization step is always recommended for staining.
