It doesn't seem like a problem with the antibody, rather some issue of fixation and other processes downstream to DAB. Definitely, it is not specific staining, but there is probably some sort of water retention in the tissue maybe during the time of fixation. Also, try to use controls and different dilutions during standardization process. If you could message me here or privately, about the protocol used, I could help you further. Also, which tissue is this?
do you have an image in brightfield, not phase contrast? Is the tissue in mounting medium and coverslipped? is there a counterstain? Just trying to be sure what I am looking at. Thanks
