If you are going to make ointment product for skin, is it essential to measure cytotoxicity for human intestinal cells? If not, is it enough to measure cytotoxicity for skin (epidermal cells) only ?
In regards to your question, the answer is depend on your purposes. Mostly, it is definitely not necessary to test the cytotoxicity for human intestinal cells. You can check the toxicity for human dermal fibroblast or epidermal keratinocytes if you are going to make a topical dosage form.
For an ointment for the skin, there is probably no reason why you would check for toxicity for human intestinal cell , only check cytotoxicity of skin cell line.
Is there any specific assays to conduct cytotoxicity tests for dermal fibroblast or epidermal keratinocytes ? could I know what is the most recommended method?
You can use MTT assay to check cytotoxicity of your samples. I use Fibroblastoma cells and check cytotoxicity by using MTT assay. And one more thing if your ointment is for cosmetic use, human dermal fibroblast or epidermal keratinocytes are okey.
