You need to check the expression of the transfected gene by Immunoblot after transfection for few days until the oligodendrocytes mature. Its good to have a GFP tag as well to monitor the efficiency of transfection. Good luck!!!

What transfection technique are you using? I've observed plasmid expression lasting at least 2 weeks, and it takes maybe 5 days to get 80% of my cultures to express MBP and be largely multipolar, so it's feasible for individual cells. Do you need a particular transfection efficiency?

@ Stuart Jenkins I use Lipofectamine and hyperfect. How do you proceed? For how long do you keep cells in proliferating media while adding transfection stuff and then replace with Differentiating media?

Hi Arpita. I use magnetic nanoparticles complexed with plasmid. These are incubated with cells for 24-48 h, in chemically-defined (serum-free) proliferation medium. Transgene expression is apparent at about 4-6 h, and peaks at ~48 h post-transfection, whether cells are kept in proliferation medium or switched to differentiation medium. I have reported greater efficiency when transfecting OPCs versus oligodendrocytes.
