To answer to your question. I am not sure that this HPLC configuration is the best way for your analysis. principally The problem is due to to the detector used in this configuration. UV detector seems not appropriate and the better way is to used refractometer detector for these carbohydrates.

Really thank you very much for your kind concern and appreciable reply.
I was asking this question because here in the lab in Tokyo, this is the HPLC configuration exist. So, I have to find another facility to analyze the total reducing sugars and lignin instead of the quantitative analysis of cellulose, hemicellulose, and lignin individually.
I hope I can achieve my goal!

Sugars have only a very weak UV absorbance signal working at low wavelengths, such as 190 nm. You will detect glucose and xylose under these conditions, however, they are not recommended for quantification due to the high absorbance of impurities at 190 nm.
