SDS- PAGE may help and I also know that this will help in finding approximate molecular weight. For proper molecular weight, we have to rely on mass spectrometry. But please let me know if there is any other method (special reference is that the antibody against the unknown Protein is provided. I want to know the whole mechanism of that technique.
You can try using size exclusion chromatography in combination with protein markers and UV detection (280 nm / 214 nm) or SEC-MALS to be more precise. You will never be that accurate without mass spectrometer but I think SEC-MALS would be the best solution.
Do not forget that you protein needs to be purified before doing such analysis.

Yes, SDS-PAGE performed with molecular weight markers is a first approach but you only obtain an estimation of molecular weight. For an accurate determination you need perform MALDI-TOF MS.
