I'm preparing a pilot to evaluate ideal time course and concentration to induce oxidative stress in cell lines (NRK-49F, B16, J774) with H2O2.
For how long and in what concentration would you suggest me to do it? I'm thinking 1h, 2h, 4h and 8h and 1uM, 5uM, 10uM, 50uM and 100uM.

sound good however, I would go down than 1uM like 0.1 OR 0.5uM and 30 min for a time so on. also I recommended to use fresh H2O2 and try don't use the H2O2 after long time opening this would reduced the H2O2 activity, maximum 2 weeks use after open the H2O2 bottle. also I would validate the H2O2 activity by checking OXO-guanine lesion in the DNA.

i can´t tell you an exact answer, but we test H2O2 on MoDC´s. Look at attachment. May be it´s a clue
