Do you want to measure it by biochemical methods or by fluorescence methods ?
By biochemical methods I think HPLC or LC/MS would be methods of choice.
By fluorescence you cas use nonyl-acridine-orange (which fluoresces green).
It has big affinity for cardiolipids.
By spectrofluorimetry you standardize with NAO alone, then make a concentration curve by adding growing concentratiions of purified cardiolipids with a constant NAO concentration. In later step you could exctract mitochondria from your frozen tissue by subcellular fractionning, and then determine the concentration of cardiolipid in your mitochondria enriched fraction.
