I am trying wrapping Ecoli BL21 cell extract into POPC liposomes but they burst in a very short time. However, when I wrap LB in the liposomes, they remain stable for a long time.(Liposomes are in PBS buffer)
We suspect it is because there are still remaining lipases in our BL21 extract and these lipases degrade our POPC membrane. We do not exclude the possibility that osmotic pressure or other components in the cell extract may play a role in our liposomes' burst.
Did you come across similar situations or do you have any ideas about the burst?
