I want to use an oily adjutant for my vaccine formulation, but to confirm its sterility, I need to perform an indirect sterility test using the filtration method. In fact, my oily adjuvant passes through the filter easily, but when I try to transfer the filter to the culture medium, it breaks into pieces. It seems the oil has a bad effect on the filter. The catalog number of my filter is: HAWG047S6. On the other hand, I don't know if I need to wash the filter with a buffer after filtering the oil. Because it passes easily through the filter and after filtration I transfer the filter into culture media. If so, what buffer do you recommend? I will be so appreciative if you help me and send me a rational and clear standard operating procedure. (I have checked the USP but it is not satisfyingly for me unfortunately). Thanks Golnaz
