Did anybody actually check the pH before and after autoclave? My different medias are starting to be acidic after autoclave and I can't find an explanation for this observation?
There are less likely pH will change in media if we autoclave. Sugars like glucose are not allowed to autoclave. May be this could be because of autoclaved sugars.
Thanks for your answer, Actually my medias are SCDA, FTM, ATM and SCDM. They are ready to use. I only weight them and solve them in purified water. Before autoclave everything is OK, But after autoclave the pH is changing to acidic point and I can't understand the reason behind this observation!!
Bacillus stearothermophilus vial can be procured. This vial should be attached to the wall inside of the autoclave. After sterilization, the vial should be kept incubated at 55 C for five days. If colour changes noticed, then the autoclave is not working good.

We have autoclave label that confirms the sterilization is okay but you told something about degrading sugars inside of the autoclave. Do you think this bacteria will be a good testament for proper autoclave procedure?!!

Now I am confused 🙄The bacteria can be died if temperature reaches above 121 so it couldn't be a good way to check the proper autoclave procedure. Do you know what I am saying?
