I did the Allium cepa assay but didn't see any chromosomal abnormalities or divisions, I want tips to ensure a successful experience and What is the best dye? Acetocarmine or Basic fuchsine?
I have worked extensively and [published on on Allium cepa and A. sativum chromosomes. This is one of the best materials for studying "assay analysis". The timings of fixation and fixation duration are very important. Staining is not the problem.

The meristematic cells in root tips and pollen from anthers of flower buds of Allium divide spontaneously. In Allium assay, root tips are considered for preparation of chromosomes. For obtaining chromosomes, the cell division has to be arrested in metaphase by adding para-di-chloro-benzene (PDB) or esculine (check spelling). The concentration and duration of treatment needs to be checked and optimized. For somatic chromosomes in Allium, aceto orcein staining yields satisfactory result. You may see beautiful chromosomes in all phases of cell division and recognize aberrations comfortably. For concentration and duration of PDB or esculin treatment, I need to check my note book to guide you.
Currently, I study aberrations in human chromosomes in hematologic malignancies to a large extent also in birth defects, pubertal delay, reproductive failures and for pre-natal diagnosis.
