I have a salt sample with 0.24ug/g of vitamin B12. I want to quantify the amount of B12 in the sample. This is too small to be detected by UV-Vis Spec and the high salt concentration and other lipophilic component of the salt is affect the column and ionization when I tried to use LC-MS. Kindly give advice on how to desalt, preconcentrate before analyzing.
if the sample in in 100% water, pass it thru an 60 mg Oasis HLB (not condition needed) and rinse the salt with water. B12 should stick to the SPE and elute with strong solvent (methanol or mixture of methanol:water)

After desalting or even perhaps without, have you considered the use of plain HPLC, using a UV/Visible detector at 214 nm, Luna 5 µm C18 Column and a 20 mM Potassium phosphate/acetonitrile (95/5 v/v) mobile phase.
