If you are limited by Raman method, you will need a micro Raman instrument: a combine of an AFM and Raman spectroscometer. In this system AFM cantilever is used to save the same distance between tip and surface, or, in other wolds, to save a focus distance for laser exciting Raman response. In this case it is possible to get a 3-D Raman signal distribution of a top surface layer by using a special program. See in attached image: 3-D distribution of GeO2 Raman signal on porous Ge surface.
