First I tried taking the absorbance for the drug release studies. But the values were abrupt. So now I am taking the fluorescence value. But the spectra collected in case of standard and drug released samples is different. I have taken fluorescence for standard curve at pH 4 and 7 just like the test samples. So can anyone suggest me what shall be done?
Are you sure no more components of the formulation are not present in the analysis sample? It is possible that interference could be a consequence of the presence of some pharmaceutical aids.

Pranjali Yadav, it is likely that your molecule aggregate and as a result suffer changes in the spectra and in the quantum yield. Is this done in water? I recommend that you perform your standard curve and sample analysis in surfactant solutions. Excess of surfactant is very likely to decrease aggregation, see the paper:
