The micellization of beta casein would occur in the condition described below, then we can calculate the CMC of protein by pyrene.

The experiment conditions in my work were: 1.5 mg of pyrene dissolved in 1 ml ethanol, then diluted in phosphate buffer to give 1.5 * 10^-6 M of pyrene, beta casein stock = 30 mg/ml, pH 7.4, T = 37 C

However, the I1/I3 ratio of pyrene-beta casein interaction in my experiment was lower than 1. Whereas, in literature, this ratio was higher than 1.

How I can fix this problem?
