i have plant extracts from different type with in the same species and i used in folin test but some of them appear that have a peak on 725 nm with absorbance equal 4 others give 250 nm approximately with noise
what is the error? i triplicate each sample but the extract is a little and i can't do many further samples again?
how i can optimise the conditions and get better results? and where is please my errors?
i do the test under shade because the materials sensitive to light
Start from a single drop in a cuvette filled with distilled water and go ahead
Full fill the cuvette with water and added one drop of NPs sample, then record the UV. If not then increase the NPs concentrations.
If I understand your problem correctly, then you have an absorption of about 4? This means you have a too high concentration of the plant sample. You should dilute the samples about 10 times, so that the absorption is below 1. In this case, do not pay attention to the absorption at 250 nm.
