Recently I conducted the small RNA sequencing of extracellular vesicles (EV) from a donor cell line and found several highly expressed miRNAs. Therefore, I tried to check the expression of them in the recipient cells after 24-hour incorporation with those EVs by qPCR. However, I could not detect some miRNAs (even though they express highly in EV). I assumed that this cell line might not express those miRNAs (I have checked on the database), and the result in qPCR might be the expression of miRNAs of the recipient cell, not the miRNAs profile from EV.

Due to my limited background in miRNAs and EV, I would like to hear suggestions from experts and anyone having experience in this field. Thank you very much for your time.
