I am selecting my transgenic seedlings on soil by spraying them with a 0.1% Challenge solution. Anybody who had similar problems?

i too had a problem with this vector but in my case i could get the resistant transgenic plants but the YFP signal was almost absent or negligible. Although a friend of mine used this vector with gene gun (bombardment method) in the onion peel and got good results.
Yes, we had that problem too, with another vector. When doing transient expression it worked fine, but when stabely transformed in plants in did not work that well. I do not know what is the reason for this.
Hi, I wonder if the YFP is the problem... this is not the first time I hear about lack of expression, with different vectors.
As to selection of transgenic lines, I really recommend you watering with basta (150mg/l in arabidopsis, I haven't tested other plants). Let the seeds germinate on soil, and when they open the cotyledons just water them with basta solution. Much simpler and cleaner than spraying, and you prevent damaging the fragile seedlings with the force of the spray, as well as prevent fungal growth. In my experience with arabidopsis, within two weeks (in short day, I haven't tried long day but it should work faster) you start seeing seedlings that are greener, and within three weeks you should clearly see resistant plants.
