Hi Antoni,

You have to follow exact protocol for southern blot analysis of the bacterial samples...It is not clear what eaxctly you are doing...is it radioactivity based detection or non-radioactivity based.....the first protocols are very straightforward..while the later ones are complex with several steps and anything can go wrong...so please elaboarte the experimental details....so that you may get good replies to rectify the problem...
