For standard curve, the cholesterol was dissolved in chloroform and then reacted with LB reagent and was fruitful. But when cholesterol was dissolved in acetone and reacted with LB reagent, an exothermic reaction was observed with pale greenish appearance. Further more cholesterol was dissolved in DMSO and reacted with LB reagent, this time no apparent changes in reaction mixture were observed. Another problem I am facing is chlorophyll in chloroform extracts. I think the chlorophyll content will interfere with final results of the samples, what do you think?
