Usually they need extra growth factors such as FGF and .... However, if your are using human chondrocytes you can use human serum and the result is amazing. I always separate serum from the donor or myself blood by centrifugation, and then decomplement at 55 C for 30 min. You can use any species own serum by collecting their blood and separate serum as well. Be aware of which donor your are collecting the blood for drug utilization and diseases and also consider 6 hours fasting before the veno-injection.
We culture primary rat chondrocytes with DMEM (low glucose) + 10% bovine fetal serum and they grow normally (see picture), but it would be great if you could use a chondrogenic medium
We manufacture chondrocytes at StemBioSys. We culture in aMEM +10% FBS and they grow well. We also use an extracellular matrix to help reduce dedifferentiation during culture.
