I want to visualize the bacterial colonies grown inside a closed system made of transparent polymer. For this, I am planning to use inverted fluorescent microscope. Please suggest what magnification will be best 40X or 100X? Also, kindly share the fluorescent dyes to be used to stain bacterial membranes?

Hi Anuj,
Magnification depends a bit on what you want to follow exactly. If you only need to follow the growth of colonies, 40x objective seems to be enough, as Michael Uebel said.
About fluorescence, make sure your polymer does not autofluoresce, it will give you some noise. I don't know how you really want to observe that, but you might need a large field of view (both for the camera and for the microscope).
