please carryout trials with full strength MS + different concentrations(0.5, 0.1, 1.5 mg/L) of indole butyric acids (IBA). this is a plant growth regulator reported in many tissue culture studies to promote root induction. However its effect vary with concentration.
Hi, I would suggest you to grow these plantlets for a while and let them be healthier and stronger with at least 2-3 unfolded leaves, and then make some wounding cuts at the basal stem and then treat the plant bases with IBA put them into media supplemented with IBA at suitable conc. Please be noted that IBA is not water soluble, so dip the stem in IBA ehtanol solution. Or use DMSO to disolve IBA first and then add to the media. GOod luck. This plants shood be able to root for they can be propagated using cuttings.
thank you so much every one for your kind suggestions.i actually tried IAA, 2,4 D & NAA. but recently i found callus formation at the base of 6 cm long in vitro grown shoot & wilting of that shoot, while i transferred it to rotting medium. what would be the reason behind?
