I am working on minicells loaded with the drug, I want to test the antimicrobial activity of the drug loaded in minicells using agar diffusion method, many trials didn't produce any zone of inhibition it may be due to problems in drug release , so what l can do?
Mini-cells are bacterially derived nano-sized particles employed in the packaging of especially chemotherapeutic drugs, for targeted delivery. It could be problems with stability, or the preparation of the cell itself. Intended application could also be a problem, not to mention the quantity and quality of agar. There are obviously many factors, so please clarify your methodology and your objective. Ensure that the cell is properly developed. Its a new technology which requires very gradual and careful optimization. Please read the attached document
Thanks Dr. Maurice for your answer , l am working on antibiotics loaded in minicells, the generated minicells were stable, the loading was performed by rotation using shaker incubater at different time intervals, then the drug was tested in the Mulleur Hinton agar but unfortunately there is no zone of inhibition in the minicells loaded with drug so the problem may be due to drug release.
Is there is any suggestion for drug release?
Why do we extract drug from minicells to release drug, the drug should be released from the drug delivery system to ensure that its successful carrier.
