I am trying to release antibodies (C225/Cetuximab/Erbitux) that have been physisorbed to the surface of PLGA-PEG-MAL nanoparticles without causing them to denature irreversibly. I am considering changing salt concentrations but am not sure about a protocol and would like some advice. Help is greatly appreciated!

There are many ways to change the thermodynamic state of an adsorbed system induce desorption: salts, pH, temperature, or solvents could work; however, it may be system specific. Below is an article which may help guide your work

Thank you Nolan. The paper definitely helped me understand the interface behavior and suggests changing temperature would induce some desorption, but maybe not to the extent I am looking for.
Are there protocol recommendations for other methods such as pH, salts, and solvent replacement out there? Thank you.

I think that you should first examine the effect of pH on the sorption of antibody on the surface, to determine that at what pH sorption of antibody is at lowest level? Then check the desorption process in that pH. The same procedure applies to the study of the influence of ionic strength.
Thank you for your response Maryam. I feel as though pHs deviating from the norm are very likely to denature the protein, does there tend to be a larger safe range than I understood? Thanks
The proteins are often split up into two groups. They are called soft protein, which have a low internal stability and the others are called hard proteins, which have a high internal stability. You should see that the protein has been used in your work, among which group of proteins?
Although protein adsorption is usually only partial reversible because proteins undergo structural changes due adsorption and they get attached with many segments to the surface. But, Changing pH or increasing ionic strength can desorb proteins. You should see that what are the isoelectric point of nanoparticles and protein have been used in your work?
