I want to ask that if we are going to check the antibacterial effect of NP's encapsulated with the drug through disk diffusion method then how we can make a disc of NP's encapsulated with the drug? Is it necessary to add any diluent or binder to make the compacted disc.

The following link contains You tube that describes a procedure for the formation of zone using disc diffusion. Zinc Oxide nanoparticles was used and tested its antimicrobial activity against S.aureus bacteria.
You can follow the same procedure for testing your antibacterial NPs.

Still I am confused because I am going to use NP's encapsulated wiith the drug where drug release pattern is very important and in case of ZnO NP's it is not so important. In many papers, I have seen that the authors make a disc of NP's encapsulated with drug but how? I don't know...
