How we can keep same amount of DMSO in all serial dilutions when we are using "2-fold serial drug dilution method" by transferring double amount in first test tube or microtiter well then taking half amount from this & transferring to next, & doing same for all required dilutions?
But this created unequal DMSO amount (DMSO amount decrease by half in subsequent next dilution).

If you had to. Then a DMSO control of the same concentration should be used
The alternative method is to make a different concentration of the compound directly separately while maintaining the DMSO concentration>

For AST what we do is get the drug in the end concentration of DMSO (lets say 10%) and make the dilution series in 10% DMSO to keep to final DMSO concentration constant throughout the series. Hope this is helpful.
