Please suggest, How to use colloidal coomassie stained gel for western blotting? Will the dye interfere with transfer? or after transfer if I destain the blot for few second in methanol, will it interfere with antibody binding?
With colloidal coomassie, you should theoretically be able to just transfer with no modification of your stained gel. It is possible that you might see some change in your protein transfer, but you should determine that empirically.
You can, however, coomassie your membrane after doing your immunoblot to use as a loading control: http://pubs.acs.org/doi/abs/10.1021/pr1011476
