I hope you have isolated pure colonies on an agar plate. Within a LAF, with the flame on, hands sanitized using an alcohol based sanitizer, hold the plate in your left hand while slightly sliding the plate's lid to open (to be done within 10-12 cm from the flame). Heat an inoculation loop until red hot; wave it in the flame's vicinity to cool it, a little. It shouldn't make a hissing noise when you are touching your plate; cool it until then. Focus to a region on the plate where you are able to see single colonies; pick just one from your plate. Now, unplug your flask containing the sterile liquid media (lb or nb) in the flame's vicinity, then dip your loop once. Heat the mouth of your flask for a few seconds and plug it off. Heat the loop until red hot, cool it. Wipe your hands again using sanitizer and incubate your flask until the required growth is achieved.
It is very easy to transfer bacteria from agar plate to liquid medium.You pick up the single pure colony from solid medium by the help of sterilized inoculation loop and transfer into the broth ( liquid medium) and incubate at room temperature for growth of bacteria.
