I am studying the NK cells as well. Previously I use lipo 3000 and lipo RNAiMAX to transfect the siRNA into NK cell. However, it seems useless. The siRNA almost cannot enter into NK cell. So I want to ask how you transfect the siRNA.
It has been discussed many times already on the forum.
Lymphocytes (B, T or NK) are simply not amenable to transfection by using chemical reagents (working through the endocytosis pathway).
FACS data show that uptake is actually achieved but once into the cells there is no further processing to induce silencing. To my knowledge there is no proofs of exact mechanisms but it can been assumed that some degradation of the transfection complex occurs...
The only way is to use electroporation or viral transduction.
