Hello people,

I am using Bio-Rad TC 20 automated cell counter to count my nuclei. I took 2ul of my nuclei suspension and added 8ul water (1:5 dilution) and further added 10 ul Tryphan blue (1:1 dilution). Since the cell counter already takes the Tryphan blue dilution into account, I thought of simply multiplying the final value given by the cell counter with the water dilution factor which is 5. However, my colleague is suggesting that I should multiply the number given by the cell counter with 2.5 ( water dilution/Tryphan blue dilution). Can someone tell me how to proceed with this? Let's assume the total count given by the machine is 4 million.
