Is it possible to store OPD after mixing with buffer ( Phosphate-citrate buffer ) and use it later as a substrate by adding H2O2 ?
Will it deteriorate the properties of OPD and affect the sensitivity of ELISA ?

Your question is of great interest and curiosity. Nevertheless, given the delicacy with which OPD alone is handled, both in usage and storage, I do not think it would be a good idea storing it in mixture with buffer. Again, buffers are not known to be stable either, especially in-house prepared buffers. Thus, for the sake of obtaining quality results, immediate usage of the mixture is advised and whatever is left of it should preferably be discarded.
